Tocotrienol As A Potential Anticancer Agent
Vitamin E is composed of two structurally similar compounds: tocopherols (TPs) and tocotrienols (T3). A growing body of evidence suggests that in addition to its antioxidative and pro-apoptotic functions, T3 possesses a number of anticancer properties that make it superior to TP. These include the inhibition of epithelial-to-mesenchymal transitions, the suppression of vascular endothelial growth factor tumor angiogenic pathway and the induction of antitumor immunity. More recently, T3 has been shown to have chemosensitization and anti-cancer stem cell effects.

Tocotrienols: Inflammation and Cancer
Inflammation has long been associated with the development of cancer. The distinct characteristics of cancer cells to proliferate, metastasize, evade apoptotic signals, and develop chemoresistance have been linked to the inflammatory response. Due to the involvement of multiple genes and various pathways, current drugs that target single genes have not been effective. Natural products target multiple genes and therefore have better success compared to drugs. Tocotrienols, the potent isoforms of vitamin E, are such a natural product.

Tocotrienol-Rich Fraction of Palm Oil Induces Cell Cycle Arrest and Apoptosis Selectively in Human Prostate Cancer Cells
Tocotrienol-rich fraction (TRF) treatment resulted in significant decreases in cell viability and colony formation in all three prostate cancer cell lines. TRF treatment resulted in significant apoptosis in all the cell lines. These results suggest that...TRF is capable of selectively inhibiting cellular proliferation and accelerating apoptotic events in prostate cancer cells. TRF offers significant promise as a chemopreventive and/or therapeutic agent against prostate cancer.

Tocotrienols are a promising class of anticancer compounds that inhibit the growth and survival of many cancer cells, including pancreatic cancer. Here, we show that tocotrienols inhibited NF-kappaB activity and the survival of human pancreatic cancer cells in vitro and in vivo. Delta-tocotrienol augmentation of gemcitabine activity in pancreatic cancer cells and tumors is associated with significant suppression of NF-kappaB activity and the expression of NF-kappaB transcriptional targets

Vitamin E delta-Tocotrienol Levels in Tumor and Pancreatic Tissue of Mice after Oral Administration
In this report, we investigated the pharmacokinetics, tumor and pancreatic tissue levels, and toxicity of delta-tocotrienol in mice because of its anti-tumor activity against pancreatic cancer. This concentration was observed with the oral dose (100 mg/kg) of delta-tocotrienol which inhibited tumor growth by 80% in our previous studies. Interestingly, delta-tocotrienol was 10-fold more concentrated in the pancreas than in the tumor. We observed no toxicity due to delta-tocotrienol.

Gamma Tocotrienol Protects Against Radiation Damage
Excessive radiation exposure damages DNA, especially DNA relating to the system in our bone marrow that produces all the red and white blood cells that are vital for survival. In the current experiment mice were exposed to non-lethal amounts of whole body radiation. Stem cell colonies were 80% - 86% maintained in the gamma tocotrienol treated mice, while they were 50% reduced in controls. A detailed analysis of the bone marrow showed that gamma tocotrienol maintained the regenerative integrity of bone marrow cells.
